Details of work done so far and significance of the scientific contribution
A) National Postdoctoral Fellow:
Currently as a postdoc fellow in a SERB-DST sponsored poject entitled “Phytochemical analysis antidiabetic mode of action of Portulaca oleraceae and Atriplex Hortensis; the two ignored crops of Kashmir region” under the mentorship of Dr, Imtiyaz Murtaza, Assoc. Professor (Biochem)/ Sr. Scientist, Division of Basic Sciences and Humanities in the Biochemistry and Molecular Biotechnology Laboratory, SKUAST-K. As postdoctoral researcher and principal investigator of this project, the prime aim of my research is to exploit the A. Hortensis / P. oleraceae, the two underutilised crops of Kashmir valley for the development of an antioxidant rich and antidiabetic functional food. They have been traditionally used as a folk medicine against diabetes in Kashmir region, however, the scientific for their medicinal activities is far the most part lacking.In this study, ten chemotypes of A. Hortensis and five chemotypes of P. oleraceae collected from five different locations of Kashmir region were included. Our findings showed that these ignored crops of Kashmir valley are nutritionally quite beneficial containing significant levels of essential minerals, fatty acid, vitamins, amino acid and other nutrients. They are also loaded with significant antioxidants (in terms of phenols) and other health promoting secondary metabolites including kaemferol, L-DOPA, dopamine, epinephrine, nor-epinephrine, quercetin, GABA and stigmasterol. The comparative analysis of these nutrients and phytochemicals in the selected Wasta Hakh (Atriplex Hortensis) and Nunner (Portulaca oleraceae) was also carried out. The determination of antidiabetic activity (in terms of α- amylase, α-glucosidase and invertase ) of plant extracts under in vitro conditions revealed encouraging results. These in vitro results provide pharmacological basis for the use of these crops in the management of diabetes and other oxidative stress related disorders. In line with this, these results are currently being revalidated under in vivo experiments using wistar rats as an experimental diabetic model. Hence with the above interesting findings, we are trying keenly to find out the effect of these plant extracts on downstream target molecules of insulin signaling pathway.
I hope these findings will soon appear in a good impact reputed journal as the elucidation of molecular mechanisms underlying the antidiabetic potential of such traditional edible medicinal plants will be of great significance.
B) Major research findings during Ph.D. and significance of the scientific contribution:
 A patent entitled “A novel process for development of phenolic enriched fenugreek sprouts with improved antidiabetic potential” was filed with Indian patent office(IPO) bearing application no. 3841/ DEL/2014 and this was the major contribution out of this research work. The patent is likely to be granted by the end of the current year (22nd December 2017)
 For the first time, an HPTLC based method was developed for the simultaneous determination and quantification of diosgenin and quercetin which can be used as quality control and quantification of these marker compounds in various plant samples, extracts and market formulations (Published in ISRN Chromatography, 2014).
 For the first time, germinating fenugreek sprouts were investigated for overall change in their total phenols, antioxidant potential, antidiabetic activity (in terms of inhibition of α- amylase, α-glucosidase and invertase inhibition) and rare phytochemicals (in terms of trigonelline, diosgenin and quercetin) at different stages of their germination. Our results showed that the aqueous extract of germinating sprouts observed a time dependent increase in their total phenols, total antioxidant activity and anti diabetic potential. Among the rare phytochemicals, diosgenin and trigonelline content of germinating fenugreek sprouts, however, were observed to decrease in due course of ten days germination period, in contrast to quercetin. The optimal day for possessing the enhanced levels of these metabolites was proved to be 4th day of germination. Therefore, through this investigation, a strong in vitro evidence was provided for confirmation of 4th germinated fenugreek sprouts as an excellent antidiabetic remedy (Published in International Journal Of Scientific Progress And Research, 2015).
 The results obtained from in vitro studies were further revalidated under in vivo conditions using streptozotocin indued diabetic rats as an experimental model. Our results demonstrated that aqueous fenugreek sprout extract is devoid of any toxicity and exibit significant hypoglycemic activity against the diabetic state induced by streptozotocin, but the intensity of the hypoglycemic effect varied from dose to dose. The best recognized dose was observed to be 300mg/kg bw and was comparable to that of standard drug, voglibiose. The extract treatment also normalized the various biochemical parameters (including body weight, liver glycogen, serum cholesterol, serum TG’s and serum insulin levels) within 21 days of experimental period. The extract treatment also restored the expression of various deregulated insulin signaling pathway components including AKT, GSK3β and GS, thereby creating a valuable approach to develop a functional food with enhanced diabetes preventing properties (Published in International Journal of Pharmaceutical Sciences and Research, 2016).
